Sialoglycan recognition is a common connection linking acidosis, zinc, and HMGB1 in sepsis

چکیده

Significance Sepsis is a condition wherein microbial infection leads to life-threatening systemic hyperactivation of innate immunity. Blood pH normally maintained tightly between 7.35 and 7.45, lactic acidosis with <7.3 indicates poor prognosis in sepsis, also associated low zinc levels. Release HMGB1 from activated and/or necrotic tissues plays pivotal role triggering the proinflammatory cascade late sepsis. Using an vitro whole-blood assay, we observed that cannot mediate activity at physiological concentrations. This due zinc-dependent association sialoglycoproteins, thereby preventing its binding receptors. Thus, drop concentration sepsis can release sequestered trigger inflammatory cascade.

HMGB1 redox during sepsis

During sepsis, the alarmin HMGB1 is released from tissues and promotes systemic inflammation that results in multi-organ damage, with the kidney particularly susceptible to injury. The severity of inflammation and pro-damage signaling mediated by HMGB1 appears to be dependent on the alarmin's redox state. Therefore, we examined HMGB1 redox in kidney cells during sepsis. Using intravital microsc...

Targeting HMGB1 in the treatment of sepsis.

INTRODUCTION Sepsis refers to the host's deleterious and non-resolving systemic inflammatory response to microbial infections and represents the leading cause of death in the intensive care unit. The pathogenesis of sepsis is complex, but partly mediated by a newly identified alarmin molecule, the high mobility group box 1 (HMGB1). AREAS COVERED Here we review the evidence that support extrac...

HMGB1 mediates cognitive impairment in sepsis survivors.

Severe sepsis, a syndrome that complicates infection and injury, affects 750,000 annually in the United States. The acute mortality rate is approximately 30%, but, strikingly, sepsis survivors have a significant disability burden: up to 25% of survivors are cognitively and physically impaired. To investigate the mechanisms underlying persistent cognitive impairment in sepsis survivors, here we ...

Volume, Conductivity and Scatter parameters of Neutrophils in Neonatal Sepsis – Is it a Cost-Effective Tool?

Background: Early diagnosis of neonatal sepsis is quite challenging. I/T ratio (immature to mature neutrophil ratio) is a highly sensitive marker of sepsis but is time-consuming and subjective. Off late, volume conductivity and scatter (VCS) of neutrophils are among the newer parameters available for screening a septic neonate. This study aimed to determine the correlation between the I/T ratio...

Hypercapnia and acidosis in sepsis: a double-edged sword?

Acute respiratory distress syndrome is a devastating disease that causes substantial morbidity and mortality. Mechanical ventilation can worsen lung injury, whereas ventilatory strategies that reduce lung stretch, resulting in a "permissive" hypercapnic acidosis (HCA), improve outcome. HCA directly reduces nonsepsis-induced lung injury in preclinical models and, therefore, has therapeutic poten...
